Projection and axial assessment

Segmentation uses the corrected brightfield median. The same display range is used for median, minimum, maximum, and individual planes; variation has its own range. Minimum and maximum views expose features suppressed or exaggerated by a median. The variation image includes focus-dependent contrast as well as any movement.

A z-stack changes focal depth and acquisition time together. Disappearance across planes therefore does not establish that a cell is floating. These short stacks do not establish the substrate or capture the full height of an upright rod. Per-object contrast profiles are diagnostic; attachment remains unverified.

WGA is assessed per plane against a local background; its maximum projection is used for visual screening only. Top/bottom localization remains unresolved.
